17. (a) Describe any two methods to force an alien or recombinant DNA into a host cell.
      (b) A recombinant DNA constructed with human DNA was introduced into a bacterial cell, but the transgenic cell did not produce the desired protein. What in your opinion could be the reason for it?

A) Two methods by which an alien or recombinant DNA is forced in Host cell are -

a) Microinjection - Foreign DNA is incorporated into the host living cell by using micro pipettes. Tip of the glass pipette is heated until it resembles a needle. The recombinant DNA is transferred from the Tip of the micro pipette to the Cytoplasm of the host cell under a powerful microscope.

b) Biolistic or Gene Gun - The target cells are bombarded with DNA coated with gold particles. Gases such as Helium or High voltage electronic discharges drives this process.

B) Mammalian recombinant DNA contains Exons an Introns. As Bacteria do not have any machinery to splice out Introns from the primary transcript , it becomes difficult to produce any desired protein from the transcript.
