p53 on the other hand activates transcription of p21 after DNA damage. 
Mechanism -

DNA damage results in activation of p53. 
This in turn activates p21
This leads to cell cycle arrest. 
This also helps in DNA repair and apoptosis.
Thereby maintaining genomic integrity.

Cancer or tumor formation is a probable clinical condition that may arise if p21 and p53 function fails.
