DIFFERENCE BW ORI AND CLONING SITE IN CLONING VECTORS 

Origin of replication (ORI) refers to the specific sequence of nucleotide present in the DNA from where the replication starts.So, ORI site is responsible for the autonomous replication of the cloning vector. 

Cloning site refers to the specific sequence of the nucleotides present in the DNA, where foreign DNA is inserted. The cloning site of the vector contains restriction sites for various restriction enzymes, such that the vector can be cut open by the enzyme and DNA can be inserted into it.
