Mature insulin consists of only two peptide chains − A and B that are linked by disulphide bonds. This mature insulin is a functional hormone. It helps in the uptake of glucose from blood and maintains its optimum level. 
In mammals including humans, Insulin is produced as pro hormone that consists of 3 peptide chains − A, B, and C. This pro hormone needs to be processed before it becomes fully mature (functional), so that it can be used to cure diabetes in humans. 
It is done by removing the extra stretch of peptide i.e C peptide from the pro hormone.

Scientists of Eli Lilly company of America prepared two DNA strands similar to the DNA strands that produce A and B peptide chains naturally in mammals by rDNA (recombinant DNA) technology. These DNA strands were then introduced into the plasmid of E. coli, where the transcription and translation of these two strands of DNA resulted in the formation of A and B peptide chains separately . These two chains were then separated  and combined by creating disulphide bonds and this way mature human insulin was formed without C peptide.
