If a person is climbing a ladder then the work done by ladder is 0 as the person is applying force. This is the correct statement but my doubt ia ladder is also applying equal and opposite force on the boy as normal reaction in the direction of motion, then why this isn't considered in calculating work done by ladder?

It is because the displacement of boy is due to the virtue of the force applied by the boy and not by the force applied by the ladder. In calculating the work done only that force is considered which actually causes the displacement not all other forces acting on the body.

work is said to be done if  and only of the body is displaced from its initial position. 
but here in this case the boy or the ladder do not doing any work .but  yes, the boy is doing the work done against the gravity  which does not matter in this case .

But if the boy is climbing up, he is displacing in the direction of normal reaction applied by the ladder on him
