ELISA is a biochemical technique used mainly in immunology to detect the presence of an antibody or an antigen like drug or poison in a sample. There are two main variations on this method: you can determine how much antibody is in a sample, or you can determine how muchantigen is bound by an antibody.

Procedure for ELISA technique:

In general;

An unknown amount of the antigen is affixed to the surface of the microtitre plates.

A specific primary antibody is washed over the surface so that it can bind to the antigen.

The secondary antibody that is linked to an enzyme such as peroxidase is added in the wells and finally

A substance such as chromogenic substance is added that enzyme can convert to some detectable signal.  The change in the colour of the sample in the wells indicates the positive result.
