The technique used to separate and isolate DNA fragments is called gel electrophoresis. It is based on their negative charged nature. It involves the following steps:
  • The DNA fragments are put in a gel matrix, commonly the agarose gel matrix and are then subjected to an electric field. 
  • The negatively charged molecules move towards positive electrode. Agarose gel matrix has varying pore sizes. The smallest DNA fragments move the longest distance and reach the other end while the large DNA fragments move lesser distance.
  • The gel is died with ethidium bromide to visualise the DNA fragments and the pieces of gel containing the fragments are cut out.
  • The pieces are extracted fro the gel by a process called elution and now are obtained in purified forms.
