4. Technique used to separate fragments of DNA would be agarose gel electrophoresis.

Gel electrophoresis is a powerful technique that helps to purify and separate molecules like DNA contained in a mixture and to estimate their size. The separation of the fragments is an important step of the recombinant DNA technology. The fragments of DNA separated, are analysed for the desired gene which we need to construct the recombinant DNA. Thus gel electrophoresis helps us to separate the desired gene or fragment of DNA from the mixture or unwanted DNA that is used then to construct the recombinant DNA.

5. It is not possible for an alien DNA to become part of chromosome anywhere along its length and replicate normally
because:
a. Alien DNA requires specific sequence called recognition sites (sites where restriction enzymes cut DNA) to ligate itself with host chromosome.
b. Also, this sequence should be close to the origin of replication (Ori). It is the sequence where DNA replication starts. This site is necessary for the binding of DNA polymerase to start replication. As this site may not be present in all alien DNA molecules, so an alien piece of DNA cannot replicate normally by attaching to any DNA.
