As light intensity increases, so does the rate of photosynthesis, up until a point.

When the temperature rises the rate of photosynthesis rises also. There is an optimum temperature  at which the rate of photosynthesis is maximum. Beyond this temperature, the reaction quickly comes to a halt.

Ice cold water will hamper the process of photosynthesis in the immersed leaf, even if there is sufficient sunshine because the temperature is an important factor for the rate of photosynthesis.

Destarching of plant can be done in day too.
Only if you keep it in a dark cupboard for 24 to 48 hours. Actually to destarch a plant, what you need is darkness. There should be no light so that the plant cannot carry on photosynthesis, thus producing more glucose which are hence converted to starch.
