gram positive bacteria can retain the gentian violet stain as they have less lipid content in their cell walls but gram negative bacteria cannot retain the stain as they have more lipid content in their cell wall.while gram staining, the cells are to be washed in alcohol or acetone,since lipid is highly soluble in such organic solvents,so gram positive bacteria cannot retain the stainas lipids get dissoved in acetone.
