Dear student,
a) Hershey and Chase worked with bacteriophage which infects bacteria. They wanted to know whether the protein or DNA of bacteriophage enters bacteria.
b) They used two types of culture media to grow protein of bacteriophage labelled with radioactive 35S and DNA of bacteriophage labelled with radioactive 35P to identify which enters into bacteria.
c) Blender was used to remove protein coat of bacteriophage which remains attached to the bacteria and centrifuge was used to separate lighter supernatant containing protein coat from denser component.
d) They concluded that DNA is transferred from virus to bacteria and protein was not transferred. Hence, DNA is the genetic material.
