Cell wall of bacteria is made up of peptidoglycan. Peptidoglycan is a polymer, also known as murein, consisting of sugar and amino acids and gives structural strength to cell wall. The principle of gram staining also depends on this peptidoglycan layer. Gram-positive bacteria possess a thick peptidoglycan layer, which helps in retaining crystal violet, Gram-negative posses a thin peptidoglycan layer. Peptidoglycan acts as a physical barrier to the bacterial cell.
