If restriction endonucleases work only with palindromic DNA sequences,what happens when we wish to isolate a DNA sequence which is non-palindromic?

Suppose the required gene has the sequence GTAGCTAG ,what is to be done?

Restriction enzymes recognise generally the pallindromic sequences, so they have to recognize a very specific sequence in order to carry out its task. It binds to the DNA only in one specific configuration.
BtrI belongs to a rare type IIQ of restriction endonucleases, which recognise non-palindromic nucleotide sequences and cleave DNA symmetrically within them. BtrI recognises the sequence 5′-CACGTC-3′ and cuts symmetrically.
There are no such enzyme which recognise the sequence GTAGCTAG, so this sequence won't be cleaved by any restriction enzyme.
