For a double stranded DNA, concentration of adenine should be equal to concentration of thymine and guanine should be equal in amount as that of cytosine so that adenine pairs with thymine by two h-bond and guanine pairs with cytosine by three h-bond. Whereas this Chargaff’s rule is not applicable in case of single stranded DNA.
