Dear student,
to isolate genomic DNA we need to follow following steps at the very basic nature of the experiment :
- cell lysis or disintegration of plasma membrane of cell mainly done by using detergents such sodium dodecyl sulphate.
- digestion of cellular components like protein and rna by proteases and RNases
- nuclear will also get denatured or disintegrated exposing genetic material.
- centrifuge using a proper solvent. (Generally we use chilled alcohol and even without centrifugation DNA precipitates out in presence of chilled ethanol )
for further study of that isolated DNA following are the steps :
- add a suitable dye
- run on agarose gel electrophoresis to obtain DNA fragments
- DNA fragment is then isolated using blotting techniques and that fragment can be studied further.
