Mutations arising due to change in single base pair of DNA as in sickle cell anaemia are called point mutations. There is a substitution of gene in these type of mutations. For example: purine(adenine) is replaced by another purine (guanine) or purine is replaced by a pyrimidine.

Point mutations do not disturb the reading frame unlike frameshift mutations (insertion and deletion observed). They just cause a single codon change. Hence, option (3) is correct.
